Presenter's Abstract

Transcription factors, coactivators, and RNA polymerase II form biomolecular condensates at super enhancer-controlled genes in mouse embryonic stem cells. The small size of these condensates makes them difficult to study with conventional techniques. However, we overcome this limitation with single-molecule imaging in live and fixed cells to reveal structural and dynamic properties of transcription condensates. Multi-color single molecule localization microscopy allows us to dissect the chromatin architecture at transcription condensates below the diffraction limit. Furthermore, tracking of single molecules inside condensates complements these studies in live cells.

Dr. Jan Spille, Assistant Professor, Department of Physics, University of Illinois at Chicago

 

Dr. Spille is an Assistant Professor in the Department of Physics, University of Illinois at Chicago. Previously, he was a Postdoctoral Fellow at MIT. He received his undergraduate degree in Physics from the University of Wuerzburg and his PhD from the University of Bonn in Germany. He is an expert in advanced fluorescence imaging techniques. His interdisciplinary work on genome organization and biomolecular condensates in the cell nucleus is supported by the NSF, several NIH grants, and two Scialog Awards.
