Over the past decades there has been a significant growth in demand for miniaturization in all areas of modern research and development. Evoked by many exciting applications, there is a need for analytical methods which require smaller amounts of sample.
Bruker meets this
challenge with a revolutionary NMR probe design: The 1mm (and 1.7mm)
MicroProbe. It operates with disposable 1mm (resp. 1.7) capillary sample
tubes and the sample volume of 5 (resp. 30) microliters enables use of
the lowest amounts of sample to run all high resolution NMR experiments
with outstanding sensitivity.
Due to the TXI-type probe design, the
z-gradient coil and the automatic matching and tuning accessory, the 1mm
MicroProbe can be used for a wide variety of NMR experiments.
The 1mm TXI MicroProbe offers the highest 1H
mass sensitivity for RT probes which results in an up to 4 times higher
mass sensitivity than 5mm conventional probes (with respect to the same
sample amount). The 1.7mm TXI MicroProbe offers an optimal 1H and 13C mass sensitivity.
The sample tubes are transferred into the magnet by use of shuttle, very similar to the well known 5mm spinners. The tube does not need to be adjusted inside the shuttle since all adjustments are made automatically inside the probe. These shuttles may be used in Bruker-BioSpin’s automatic NMR sample changers such as the SampleCase and the SampleJet.
The MicroProbes works with discrete, disposable 1mm sample tubes, which
may be sealed after filling. The glass tubes have a plastic, v-shaped
top part attached to them, which allows for easy manual sample filling
even with volumes as small as 5 (resp. 30) micro liters. The tubes may
be equipped with a digital matrix code on the plastic top parts, which
then uniquely identifies each sample.
Even though manual sample
filling is straight forward, with the SamplePro, Tubeᵀᴹ a sample
preparation robot is available, which enables the fast, reliable and
comfortable filling of many 1mm (resp. 1.7mm) sample tubes at a time.
Here the sample tubes are arranged in the industry-standard 96 well
plate format, so that the tube racks are fully compatible with all other
laboratory or storage equipment which is capable to handle this format.
They can also be directly used in our SampleJet.
Mass Limited Samples and Reduction of Expenses for Deuterated Solvents
Due to their high mass sensitivity MicroProbes are especially well suited for the investigation of highly soluble compounds that are only available in limited amounts. This is often the case in natural products, but also in body fluids like CSF or blood and urine from rodents. The TXI-probe design allows to run the full range of experiments typically needed in biologic samples.
Needing only small amounts of deuterated solvents helps to keep the overall cost of an experiment low and will also help to get best performance, if solvent suppression is required. Therefore MicroProbes are well established in quality control of compound libraries, where only limited quantities of protonated solutions are available for NMR spectroscopy.
The small sample volume required results in short pulses that are hardly influenced by the ionic strenght of the sample, making these Probes very attractive for salty samples, like urine.
